An Instrumental Planar Chromatographic Method (HPTLC) with Automatic Multiple Development Chamber (AMD) for the cholesterol analysis in egg yolk obtained from hens fed with diet to reduce the cholesterol content, was set up and validated. The gradient to achieve the simultaneous separation, identification and quantification of cholesterol, phosphatidylcholine and triolein was established. Recovery was evaluated through a standard addition procedure with a yielding of 99,4%. Repeatability and Intermediate Repeatability resulted adequate on the ground of Media Difference analysis at a confidence limit of 95%.
