Abstract

The enzymatic hydrolysis reaction of bovine blood proteins in a batch reactor using the response surface methodology and Genetic Algorithms as optimization tools was studied. Furthermore, the effect of substrate concentration on the reaction rate using a mathematical model, which assumes substrate inhibition, was analyzed. The Alcalase ® 2.4 L is used for the hydrolysis. The p2, temperature and the enzyme-substrate ratio were analyzed in order to maximize the degree of hydrolysis for two hours of reaction, using the method of the assessment of proton release. The effect of experimental factors on the degree of hydrolysis was satisfactorily adjusted to a polynomial model with R2 = 0.93. The optimal reaction conditions were temperature 60.4 °C, p2 9 and enzyme-substrate of 10% (w/w), to achieve maximum degree of hydrolysis of 23%.
